Roberta Glass, a bored and restless Fort Lee housewife, becomes intrigued by personal classified ads titled 'Desperately Seeking Susan'. She goes to Battery Park to meet Susan after reading an ad from Jim, a rock musician with whom Susan is involved. Roberta inadvertently buys Susan's distinctive leather jacket and suffers amnesia, leading her to believe she is Susan when she inherits all of Susan's belongings. Dez, a movie projectionist and friend of Jim's tasked with looking after Susan, mistakes Roberta for Susan and falls in love with her. Meanwhile, criminal Wayne Nolan is pursuing Susan for priceless Egyptian earrings she possesses, unaware that Roberta has taken on Susan's identity. As Roberta navigates her new life with Dez, Susan tries to find the mysterious woman who stole her belongings while evading Wayne. Complications arise when Roberta's husband Gary searches for his missing wife, and Jim returns to town looking for Susan.
